Overview

We are seeking an experienced, creative and highly motivated Computational Chemist to join our multi-disciplinary, collaborative team contributing across our portfolio in the design and analysis of new therapeutic compounds.

This role, which can be based at either of Amphista’s research hubs in Cambridge, UK or BioCity Glasgow, UK is an exciting opportunity to join a fast paced, dynamic company developing first-in-class medicines to treat cancer and other areas of high unmet medical need.

 

Responsibilities include:

Working closely with experienced medicinal chemists and other drug discovery scientists within close project teams to contribute to the design and prioritisation of protein degrading molecules as well as analysing project data and building predictive design tools and models to accelerate the optimisation process. Your contributions will directly impact multiple drug discovery projects and also help grow the proprietary Amphista protein degrading platform.

 

  • Providing computational chemistry support to the Amphista portfolio of projects. This may include a range of areas from molecular modelling & docking through to cheminformatic analyses and predictive model building. Specific areas include:
    • Design and optimisation of ligands and bifunctional degraders using x-ray structural data, homology models etc.
    • Ligand identification from literature analyses or virtual screening.
    • Collation and analysis of complex, multi-parametric datasets to rationalise SAR and build predictive models in areas such as biological activity, selectivity, in vivo properties and others.
  • Working closely with medicinal chemists to provide computational solutions to significant hit identification and lead optimisation project challenges.
  • Providing computational analyses and solutions to projects via your own personal contributions as well as by working with external vendors or collaborators where appropriate.
  • Building computational tools and models with potential utility within specific projects as well as across the entire Amphista portfolio.
  • Creating tools and solutions for use by project team members as well as expert tools.
  • Contributing to the success of one or more projects as a key protect team member working closely with colleagues from biology, drug metabolism and other groups.
  • Critically analyse project data and contribute ideas that will drive projects forward.
  • Support the continuing development of Amphista’s protein degradation capabilities.
  • Maintaining an up to date literature knowledge of computational chemistry strategies and protein degradation chemistry.
  • Creating data summaries and visualisations able to communicate and display complex datasets and conclusions in intuitive and interactive ways.
  • Present your work within project teams & wider company, cross-site meetings with the potential to also present to external audiences.
  • Maintaining electronic lab notebook records and contributing to drafting of patent applications when appropriate.

 

Required attributes:

  • PhD in a computational chemistry-related discipline or equivalent experience with at least 3 years experience of hands-on delivery of computational chemistry support in a drug discovery project setting in pharma, biotech or academic drug discovery unit.
  • Experience of structure-based and ligand-based design using shape, fingerprint and pharmacophore-based methods including library design & enumeration and lead-hopping.
  • Experience of extensive SAR analysis of project data sets, calculation of ADME/Tox properties and building of models to allow property prediction.
  • Outstanding knowledge and hands on experience of using a wide range of computational tools across molecular modelling and cheminformatics. These may include MOE, Cresset, Schrodinger tools.
  • Wide experience of usage of data analysis, visualisation & pipeline tools which may include Vortex, Spotfire, Data Warrior, Pipeline Pilot, Knime.
  • Track record of successfully working in multidisciplinary settings and in a collaborative manner where your work has directly impacted project decisions and success.
  • Strong interpersonal & communication skills able to explain complex concepts in an intuitive and interactive manner to a wide range of project members and senior leaders.
  • Proven track record of delivery from your own personal project contributions.
  • Willingness to travel to and spend significant time at both Amphista research hubs in BioCity Glasgow & Cambridge, UK to work closely with project team members.
  • Must be fun to work with.

 

In addition, it would be desirable that the successful candidate has:

  • Familiarity with storing and analysing data using the Dotmatics suite of tools.
  • A working knowledge of Python.

Responsibilities include:

  • Working at the interface of R&D, operations and finance to maintain up to date portfolio view of all ongoing R&D activities to enable optimal resource utilization to meet R&D priority goals
  • Working closely with project teams to build project plans including resourcing & budgetary needs as well as delivery timelines
  • Tracking progress of projects across portfolio, working with teams to meet agreed milestones and identify & mitigate risks and potential for timeline slippage to aid decision making
  • Working with finance team to ensure project spend is aligned with budgets
  • Scenario planning and proposing innovative resourcing solutions to increase the efficiency of delivery of all projects and R&D portfolio as a whole.
  • Working with our network of external CRO and academic partners to co-ordinate activities, timings and track progress
  • Providing both project-level and portfolio-level summaries and concise reports of the status of the Amphista portfolio to senior leadership and portfolio committees to contribute to effective R&D and corporate strategic planning and decision-making
  • Creating regular summaries of project progress and resource/budget utilization to assist programme team and leadership planning
  • Presenting updates and proposed solutions to improve delivery of projects and other objectives to range of groups including project teams, R&D & executive leadership and board level

 

Required attributes:

  • Strong scientific background through bachelors degree or PhD in relevant area of chemical, biological or computaitonal sciences
  • Thorough understanding of the drug discovery process from project initiation and target validation through to candidate selection, pre-clinical and clinical entry gained from working for 3 or more years in a drug discovery/development setting, likely in a pharma or biotech environment.
  • Experience providing project management support to successful drug discovery projects achieving milestones from lead identification, candidate nomination, clinical entry and progression
  • Expert user of project management software and tools used to collate, analyse, plan and present portfolio & project status
  • Experience providing strategic input at R&D portfolio level to allow successful achievement of R&D and corporate goals
  • Outstanding communication skills with track record of successfully working with and influencing range of stakeholders including internal R&D teams, external collaborators and executive leadership
  • Experience co-ordinating projects run across multiple sites and geographies
  • Good understanding of drug discovery & development CRO landscape and ways of working to make CRO collaborations successful
  • Proven track record in delivering high business impacting programme/projects

We are seeking a number of drug discovery cell biology & screening scientists to join our new team based in our Cambridge research hub, due to open in late 2021, which will support cell biology, screening, disease model assessment and lead characterisation across our portfolio of projects moving towards clinical studies. A number of lab-based roles are available which will suit a range of candidates from those with extensive experience of working in a drug discovery setting through to earlier career scientists recently completed a PhD or post-doctoral studies with a strong desire to begin a career in drug discovery.

Roles and responsibilities include:

  • Supporting one or more active drug discovery projects: developing assays, characterising hit and lead molecules, contributing to discussions on project planning and strategy.

  • Developing relevant, novel and innovative cell-based and biochemical assays to address project challenges and support drug discovery projects in the area of oncology as well as applying these assays to screen and profile cellular activity of novel compounds and to interrogate target biology/disease relevant mechanisms.
  • Contributing to discussions and work on target selection and project planning.
  • Culturing human cell lines and using a range of technologies to generate new cell lines.

  • Working in an interdisciplinary team to support research strategies and project goals.
  • Working with external collaborators including CROs and academic experts to seek input and generate project data to answer specific questions.

Essential requirements:

  • D in Biology, Pharmacology, Cell biology or a related discipline or a BSc/MSc degree with significant relevant experience of leading projects to successful milestones and conclusions.
  • Strong experience in molecular and cellular biology techniques applied to the development of physiologically-relevant cellular assays and systems applied to target validation, compound characterisation and screening.
  • A practical understanding of a wide range of cellular assay methods and technologies, including mass spectrometry, fluorescence and luminescence.
  • Expertise in cloning, stable cell line generation, gene expression knockdown.
  • Deep knowledge of mechanisms of disease biology in oncology, immunology and/or CNS disease.
  • Evidence of significant scientific productivity and impact through your own personal contributions leading to projects reaching significant milestones.
  • Outstanding track record evidenced by publication and presentation record.
  • Demonstrated excellence in independently planning and executing experiments as well as critical data analysis skills.

We are seeking a creative, proactive and highly-motivated Lead Bioinformatician to join our multi-disciplinary, collaborative team of drug discoverers. In this role, you will be responsible for delivering a range of bioinformatic analyses to support our drug discovery programs ranging from target identification and mode of action studies through to biomarker identification and characterisation of the potential clinical utility of lead candidates.  As a key member of project teams, you will work across our current therapeutic drug discovery projects proactively providing input to define and solve key project questions to allow advancement of the Amphista portfolio. This is an exciting opportunity to join a fast growing and dynamic company operating at the cutting edge of protein degradation to develop first in class medicines to treat cancer and other areas of high unmet medical need.

 

Roles and responsibilities include:

  • Supporting all projects from the Amphista portfolio by provision of high-quality bioinformatic analyses and other input to help projects progress quickly to reach key milestones.
  • Contributing to identification of targets where the proprietary Amphista protein degrading approach is likely to provide high therapeutic benefit to defined cohorts of patients, showing superiority over standards of care.
  • Identification and characterisation of suitable, clinically-relevant cellular systems to build data packages around lead molecules.
  • Analysis of multi-omic data sources in order to build strong supporting datasets around mode of action and selectivity of lead candidates including biomarker strategies.
  • Analysis of a wide variety of bioinformatic data sources including transcriptomics, proteomics, biological network analysis.
  • Working as part of small project teams in an interdisciplinary environment to support research strategies and project goals.

Essential requirements:

  • PhD in computational biology, genetics or a related discipline with a minimum of 1 year experience working in a multi-disciplinary drug discovery project team setting likely within a pharma or biotech environment.
  • Extensive experience in analysis of large-scale omic data (including genomics, transcriptomics, epigenetics, proteomics) using bioinformatics tools and pipelines including R, Python or similar.
  • Experience in analysing next generation sequencing data & pooled functional genomic screening data (e.g. shRNA, CRISPR) from a range of sources.
  • Accomplished user of medical and biological databases to seek insight into mechanisms of disease
  • Wide experience of applying statistical principles to biological datasets to aid interpretation
  • Good appreciation of where a range of bioinformatic analyses can have the biggest impact on advancing the drug discovery process
  • Ability to work independently, with strong initiative and drive, but also as part of a team to ensure analyses are focused in optimal areas to answer most critical project questions.
  • Excellent ability to communicate through a wide range of verbal, written and electronic means

POSITION SUMMARY

Amphista Therapeutics is a fast-growing biotech and the leading Europe-based Targeted Protein Degradation (TPD) company. Initially founded from the laboratory of Professor Alessio Ciulli (University of Dundee) our team comprises both academic and drug discovery pioneers of the TPD field and a growing R&D team focused on advancing a portfolio of medicines to the clinic using our proprietary next generation TPD technologies. We recently completed an oversubscribed $53M funding round to allow us to advance our portfolio of projects towards the clinic and greatly strengthen the development of our underlying scientific platform.

Amphista’s approaches use completely novel mechanisms to build on all the advantages of TPD while directly addressing the emerging limitations of existing TPD strategies to expand the utility of the field. We are seeking an experienced, creative and highly motivated cell biologist to join our multi-disciplinary, collaborative team based in our new research centre in Cambridge, UK. In this role, you will oversee and personally contribute to a wide range of areas related to characterising the in vitro and in vivo profiles of lead molecules from across our drug discovery portfolio.  Working within project and functional teams, this role will support target validation, lead optimisation and preclinical development of Amphista’s therapeutic portfolio and your contributions will directly contribute to the success of the R&D strategy. You will use your experience of working within a drug discovery environment to design project strategies, experimental plans and ensure prioritisation and timely generation, analysis & sharing of key data. This is an exciting opportunity to join a fast growing and dynamic company operating at the cutting edge of protein degradation to develop first-in-class medicines to treat cancer and other areas of high unmet medical need.

 

Roles and responsibilities include:

  • Developing relevant, novel and innovative cell-based and biochemical assays to address project challenges and support drug discovery projects in the area of oncology as well as
  • Applying these assays to screen and profile cellular activity of novel compounds and to interrogate target biology/disease relevant mechanisms
  • Contributing to discussions and work on target selection and project planning.
  • Culturing human cell lines and using modern technologies to generate new cell lines.
  • Critically analyse project data and contribute ideas that will drive projects forward.
  • Work in an interdisciplinary team to support research strategies and project goals.
  • Working with external collaborators including CROs and academic leaders to seek input and generate project data to answer specific questions.
  • Participate in building out the research capabilities of the Biology group.

Essential requirements:

  • D in Biology, Pharmacology, Cell biology or a related discipline or a BSc/MSc degree with significant relevant experience of leading projects to successful milestones and conclusions.
  • Deep knowledge of mechanisms of disease biology in oncology, immunology and/or CNS disease.
  • Experience working as part of multi-disciplinary teams in a drug discovery setting in eg the pharmaceutical or biotechnology industry.
    • Evidence of significant scientific productivity and impact through your own personal contributions
  • Strong experience in molecular and cellular biology techniques applied to the development of physiologically-relevant cellular assays applied to target validation, compound characterisation and screening.
    • A practical understanding of a wide range of cellular assay methods and technologies, including mass spectrometry, fluorescence and luminescence
    • Expertise in cloning, stable cell line generation, gene expression knockdown
  • Outstanding track record evidenced by publication and presentation record

As a Director of Chemistry at Amphista you’ll be a key member of the leadership of the R&D team reporting directly to the CSO. The role will be critical to driving R&D and company success and you’ll have significant impact on the success of Amphista R&D through your leadership of key projects within our portfolio and contributions to the wider R&D strategy. As a key member of the R&D team, you would ideally be located with the R&D team in Scotland though for exceptional candidates, remote working from other locations may be possible.

You’ll use your extensive experience of successful project delivery and creative ways to overcome drug discovery challenges to lead and contribute to specific programmes within Amphista’s protein degrader drug discovery portfolio, having a significant impact on the success of the R&D strategy. Working to lead project and functional teams, you’ll work across hit ID, lead optimisation and preclinical development.   • Designing and delivering the medicinal chemistry and wider project strategy for one or more high priority portfolio programmes.
  • Contributing medicinal chemistry expertise across the portfolio of projects at all stages in areas such as drug molecule design & synthesis, data analysis of SAR & DMPK profiles, building of efficacy/safety data and writing of candidate selection data packages.
  • Contributing to target selection, prioritisation and advancement discussions across portfolio.
  • Designing project plans to enable fast decision-making and project progression including nomination of clinical candidates for IND/CTA-enabling studies and clinical entry.
  • Developing project resourcing plans to make most efficient use of internal & external resources.

Essential requirements:

  • Ph.D in chemistry or a related discipline or a BSc/MSc degree with significant (more than 10 years preferred) relevant pharma/biotech drug discovery experience of leading projects to successful milestones and conclusions in oncology or closely-related therapy area.
  • A deep understanding of contemporary small molecule drug discovery and development including structure-based design, property-based design, multi-parameter optimisation & successful use of computational chemistry & cheminformatic tools.
  • Experience of contributing to company or business unit R&D strategy through creative identification and prioritisation of opportunities.
  • Evidence of significant scientific productivity and impact through your own personal leadership of projects to key milestones including candidate selection and clinical entry.
  • Extensive experience of implementation of creative medicinal chemistry strategies to produce high quality candidate molecules for subsequent development and clinical evaluation.
  • Highly scientifically creative and flexible as evidenced by successful delivery of “non-standard” projects (e.g. agonists, covalent modifiers, bifunctionals/PROTACs, less common dosing regimens & routes).
